Production of Monoacylglycerides by Enzymatic Alcoholysis of Immobilized Rhizopus delemar Lipase

Abstract

Lipase (EC 3.1.1.3) from Rhizopus delemar immobilized on silica gel was used to catalyze production of monoacylglycerides (MAGs) using different enzymatic processes, hydrolysis, glycerolysis, and alcoholysis. A batch system with shaking was used with and without addition of water. In all processes, mixtures of triacylglycerides (TAGs), diacylglycerides (DAGs), and MAGs were obtained. The yield of MAGs depended strongly on the reaction temperature. At a higher temperature (50¡É), approximately 30.0% MAGs were produced at equilibrium. At a lower temperature (37¡É), yields of 88.0¡90.0% MAGs were obtained for the two oils examined. Only the oil-alcohol ratio showed no significant effect among the factors that affect the yield of MAGs, which include temperature, incubation time, and water content. The results reported here clearly show that a high yield of up to 90.0% MAGs may be obtained using different oils by immobilized lipase catalyzed alcoholysis employing non-aqueous system. Thin layer chromatography (TLC) and high pressure liquid chromatography (HPLC) were used to identify and quantify the products formed. Mixtures of MAGs (monolaurin and monomyristin) were detected.
